Direct proof of use
The S-400 Triumf was fielded by Russian forces in Syria during the Syrian Civil War. RUSI reported that Russia deployed the system to Latakia air base after a Turkish F-16 shot down a Russian Su-24 near the Syrian border in November 2015, and Sky News reported Russian Defense Ministry statements that the S-400 had been sent to Khmeimim airbase near Latakia and placed on air-defense combat duty.
The documented role was long-range air defense, airspace denial, deterrence, and protection of Russian military infrastructure and Syrian-aligned assets. Open reporting does not establish routine S-400 missile firings in Syria; later analysis focused on why the system did not engage U.S. Tomahawk cruise missiles during the April 2017 Shayrat strike.

Narrative
The S-400 deployment formed part of Russia's wider intervention in support of the Syrian government. Its presence at Latakia gave Russian forces a high-profile air-defense umbrella over the coastal base area and advertised a threat to aircraft operating over much of western Syria, the eastern Mediterranean, and southern Turkey.
RUSI described the system and associated Su-35 fighters as a major problem for the U.S.-led coalition, but also noted that using S-400 missiles against coalition aircraft would carry major escalation risks. The system therefore functioned primarily as a deterrent and air-denial asset, making coalition planners account for Russian coverage without producing a publicly documented pattern of S-400 intercepts.
After the April 2017 U.S. strike on Shayrat, analysts emphasized that low-flying cruise missiles were difficult targets at long distance because of radar-horizon limits. RUSI's 2018 commentary identified two S-400 units in Syria: one covering Khmeimim and another near Masyaf in Hama Governorate, with limited ready missile capacity and directional engagement constraints.
